不同雷公藤制剂中6个成分的含量研究

  摘   要:目的  建立測定雷公藤制剂中的6种成分(雷公藤甲素、雷公藤内酯酮、雷酚内酯、雷公藤次碱、雷公藤红素及雷公藤内酯甲)的高效液相色谱方法。方法  采用Agilent TC-C18柱(250 mm×4.6 mm,5 μm),以10 mmol/L乙酸铵水溶液-乙腈为流动相梯度洗脱,流速为0.75 ml/min,检测波长为225 nm,柱温为25 ℃。结果  6种成分的进样浓度为0.1~156.25 μg/ml范围内与峰面积积分值线性关系良好,平均回收率为98.5%(RSD=1.9%)、99.2%(RSD=1.9%)、99.5%(RSD=1.5%)、98.7%(RSD=1.2%)、98.8%(RSD=1.6%)、99.0%(RSD=1.7%)。结论  所建立的测定方法操作方便,重复性强,结果准确,可用于雷公藤及其相关产品的质量分析;不同雷公藤制剂中6个成分的含量存在差异。

  Abstract:Objective  To establish a high performance liquid chromatography method for the determination of 6 components in Tripterygium wilfordii(Triptolide,triptolide,lycopolide,Tripterygium,tripterin,and triptolide a).Methods  A Agilent TC-C18 column of (250 mm×4.6 mm,5 μm)was used.The mobile phase gradient elution was 10 mmol/L ammonium acetate aqueous solution and acetonitrile.The flow rate was 0.75 ml/min,the detection wavelength was 225 nm,and the column temperature was 25 ℃.Results  The injection concentration of the six components was 0.1~156.25 μg/ml and the peak area integral value was linear,the average recovery rate was 98.5%(RSD=1.9%),99.2%(RSD=1.9%),99.5%(RSD=1.5%),98.7%(RSD=1.2%),98.8%(RSD=1.6%),99.0%(RSD=1.7%).Conclusion  The established method is convenient, reproducible and accurate.It can be used for the quality analysis of Tripterygium wilfordii and its related products,and the content of 6 components in different Tripterygium wilfordii preparations is different.
